Neoplasia induction in NOD.CB17-Prkdcscid/J by passage of live tumor cells (HT-29)

Protocol

 

Purpose: To provide normative data for testing biologically-active compounds in the immunodeficient strain, NOD.CB17-Prkdcscid/J, which is an excellent host for xenografts. The human colorectal adenocarcinoma cell line, HT-29, is used for tumor induction.

This protocol indicates a treatment schedule. Data from these tests are proprietary; however, data are available from controls and pre-treatment metrics for mice in the Test Group.

By posting this data, JAX® In Vivo Services contributes to community research resources and fulfills an agreement with clients to make normative data public.

Experimental Groups:

Control (no vehicle) mice receive the HT29 injection only and no further treatment.

Control-vehicle: mice receive the HT29 injection followed by weekly injections of saline (vehicle; intra-peritoneal).

Test Group (treatment): Proprietary

 

Reagents

Saline (sterile)

HT29 tumor cell line (ATCC #HTB-38) suspension in saline

 

Procedure

Mice receive a single injection of up to 1x106 cells (HT29) in 100 µL, subcutaneously in the dorsal flank.

One week later, Control-Vehicle mice begin receiving weekly injections of saline, for a total of four injections.

Mice are weighed and tumor sites are palpated three times a week (measured with a digital caliper). Body weights and tumor metrics are recorded.

Tumors are harvested at the end of the study and weighed, snap frozen, or fixed in formalin.

 

Definitions & Formulas

Tumor volume = (tumor length * tumor width2) ÷ 2

 

Data

Body weights and tumor metrics were measured several times a week over the course of the experiment (see Workflow). Changes in body weights and tumor volume were determined as a percentage of prior measurements (specified in data descriptions). Per-animal data are available along with the latency (days) for tumor volume to reach 500 and 1500 mm3.
