Purpose: To measure lipid and glucose levels in non-fasting B6.129P2-Apoetm1Unc/J and B6.129S7-Ldlrtm1Her/J knockout strains from 4-40 weeks of age compared to C57BL/6J, and to assess atherosclerotic plaques at 8 and ~45 weeks of age (retired breeders).

Methods: Blood was collected from the submandibular route into serum separator tubes. Total cholesterol (CHOL), low density lipoprotein (LDL), high density lipoprotein (HDL), triglycerides (TG), glucose (GLU), and non-esterified free fatty acids (NEFA) were measured using the Synchron CX5 Delta (Beckman Coulter, Inc., Brea CA) clinical chemistry analyzer. A minimum of five mice were analyzed per sex, age, and strain. CHOL, LDL, HDL, and TG are plotted below.

Hearts and aortas were removed, fixed, and cryo-sectioned on 8-week old mice and retired breeders (approximately 45 weeks of age). Slides were stained with Oil Red O. Representative sections are below.

Pathology

 

Pathologist's comments:

  1. Apoe- and Ldlr-null mice have atherosclerotic plaques at the base of the aorta at retirement age, but the plaques are larger in Apo-null mice.
  2. All of the Apoe-null mice have significant plaques throughout the rest of the aorta, while none of the Ldlr-null mice have grossly detectable plaques.
  3. None of the strains had plaques at 8 weeks of age.
  4. No significant differences were noted between males and females, although there may be slight differences that would necessitate large numbers of mice and/or quantitative analysis to be statistically significant.
